Seeing the small with lightsheet fluorescence microscope
Have you ever wondered how light can help doctors in disease diagnosis? Light microscopy is a great imaging tool that can monitor diseases like cancer without doing a biopsy in clinics. You will get to characterize our lightsheet microscope, image different samples, and learn about how it helps in cancer staging.

Objectives:
- Understand the principles behind light microscopy with special focus on lightsheet microscopy, including advantages and drawbacks.
- Acquire 2D images of fluorescence from fluorescein test sample.
- Acquire 3D images of fluorescent test beads.
- Characterize the light-sheet field-of-view.
- Characterize the resolution of the microscope.
Hvordan foregår øvelsen?
On the day of the exercise you will get to use the state-of-the-art multiphoton lightsheet microscope in the biophotonic imaging group laboratory.
You will be able to do fluorescence imaging of samples like fluorescein and fluorescent beads.
You will learn more about light microscopy and why lightsheet microscopy is preferred in various applications.
You will also get to keep the imaged data for yourself, and work on different ways of visualizing the data.
Hvilke faciliteter kommer du til at bruge?
You will be introduced to the state-of-the-art bio-imaging laboratory facilities that the biophotonic imaging group at DTU Health Tech offers. The exercises will be done in the same lab.
You can use the data from the exercise to understand how to characterize a lightsheet microscope in terms of its field-of-view and resolution.
You will learn what imaging parameters to look for specific to applications.
The theory will be based on optics and microscopy.
Tentative schedule:
Day 1: Intro to lab safety & lightsheet microscope setup, 2D imaging of liquid fluorescein sample, discuss next day's work step by step.
Day 2: Repeat safety instructions, lab walkthrough of setup, 2D imaging of microbeads, data analysis.
You will acquire the 2D images of fluorescein and fluorescent beads using the multiphoton lightsheet fluorescence microscope.
You will take home the image files that you acquired during the exercise. During the exercise, we will also discuss how to characterize the field-of-view and resolution of the lightsheet microscope.
You may take home the characterization data as well.
You will recieve detailed instructions on how to operate the lightsheet microscope and the safety constraints of using the femtosecond laser.
I would be preferable if you go through basics of how a fluorescence microscope works. A good literature overview is present in chapters 3, 4, 10, and 15 of the textbook "Understanding Light Microscopy" authored by Jeremy Sanderson (DOI: 10.1002/9781118696736).
